Now, research from the University of Pennsylvania, in collaboration with Indiana University, suggests that an FDA-approved therapy for iron-deficiency anemia also holds promise for treating, preventing, and even diagnosing dental decay. The therapeutic, a combination of an iron-oxide nanoparticle-containing solution called ferumoxytol and hydrogen peroxide, was applied to real tooth enamel placed in a denture-like appliance and worn by the study subjects.

The study, published in the journal Nano Letters, found that a twice daily application of ferumoxytol, which activated hydrogen peroxide contained in a follow-up rinse, significantly reduced the buildup of harmful dental plaque and had a targeted effect on the bacteria largely responsible for tooth decay. These types of nanoparticles with enzyme-like properties are sometimes known as ?nanozymes? and are increasingly being explored for their potential in biomedical and environmental applications.

?We found that this approach is both precise and effective,? says Hyun (Michel) Koo, a professor in the University of Pennsylvania School of Dental Medicine. ?It disrupts biofilms, particularly those formed by Streptococcus mutans, which cause caries, and it also reduced the extent of enamel decay. This is the first study we know of done in a clinical setting that demonstrates the therapeutic value of nanozymes against an infectious disease.?

The work is an extension of a 2018 paper published in Nature Communications, in which Koo and colleagues, including David Cormode of Penn?s Perelman School of Medicine, showed that the iron oxide nanoparticle-hydrogen peroxide treatment could prevent biofilm accumulation and tooth-decay in an experimental model and an animal model.

In the current work, the scientists wanted to take the next logical step, working in humans. In a randomized study, they had 15 participants use a removable, denture-like device with real tooth enamel attached, a method developed and extensively tested by Domenick T. Zero of Indiana University, a co-corresponding author on the current paper.

The study participants applied a sugar-containing solution to the appliance four times a day, mimicking high-sugar meals and snacks consumed in the course of daily life. Participants were asked not to brush the enamel specimens but instead to rinse the appliance twice a day. Participants were divided into three groups, with one using the ferumoxytol then the hydrogen peroxide rinse, one with a solution that provides the inactive ingredients in ferumoxytol, and a third with water alone.

After 14 days, the researchers analyzed the biofilms that accumulated on the enamel specimens. They found the experimental treatment potently reduced the growth of biofilms containing S. mutans and could kill this bacteria with high specificity. Other commensal bacteria normally found in the mouth were not affected by the ferumoxytol-hydrogen peroxide therapy.

The results and safety of the approach were supported by previous work, which has shown that the iron oxide nanoparticles do not bind to the mucosal tissue in the mouth and do not cause cytotoxicity or changes in the oral microbiome in an animal model.

?This treatment doesn?t seem to have harmful, off-target effects,? Koo says.

The reason for this precision is three-fold. The reaction to catalyze hydrogen peroxide only happens in a highly acidic environment, such as what arises when caries-causing bacteria are present and active. The treatment also causes the sticky matrix of carbohydrates to break down, degrading the biofilm and exposing the microbes. And ferumoxytol can specifically bind to receptors on the S. mutans cell membrane, facilitating its killing.

In a final stage of the study, the researchers added a marker that turns blue when exposed to reactive oxygen species, like those generated by the catalysis of hydrogen peroxide by ferumoxytol nanoparticles. And indeed, the team found that the intensity of blue labeling corresponded with acidic biofilms containing S. mutans.

Because of this ?detection? capability, Koo notes that the experimental treatment has the potential to become what is known as a ?theranostic,? that is, a drug that can be used to both diagnose a condition and treat it. ?It can also be used at home,? Koo says. ?You could rinse with it, see how much cavity-causing plaque there is, and then treat with the solution or consult a dentist for follow-up treatment.?

Now, research from the University of Pennsylvania, in collaboration with Indiana University, suggests that an FDA-approved therapy for iron-deficiency anemia also holds promise for treating, preventing, and even diagnosing dental decay. The therapeutic, a combination of an iron-oxide nanoparticle-containing solution called ferumoxytol and hydrogen peroxide, was applied to real tooth enamel placed in a denture-like appliance and worn by the study subjects.

The study, published in the journal Nano Letters, found that a twice daily application of ferumoxytol, which activated hydrogen peroxide contained in a follow-up rinse, significantly reduced the buildup of harmful dental plaque and had a targeted effect on the bacteria largely responsible for tooth decay. These types of nanoparticles with enzyme-like properties are sometimes known as ?nanozymes? and are increasingly being explored for their potential in biomedical and environmental applications.

?We found that this approach is both precise and effective,? says Hyun (Michel) Koo, a professor in the University of Pennsylvania School of Dental Medicine. ?It disrupts biofilms, particularly those formed by Streptococcus mutans, which cause caries, and it also reduced the extent of enamel decay. This is the first study we know of done in a clinical setting that demonstrates the therapeutic value of nanozymes against an infectious disease.?

The work is an extension of a 2018 paper published in Nature Communications, in which Koo and colleagues, including David Cormode of Penn?s Perelman School of Medicine, showed that the iron oxide nanoparticle-hydrogen peroxide treatment could prevent biofilm accumulation and tooth-decay in an experimental model and an animal model.

In the current work, the scientists wanted to take the next logical step, working in humans. In a randomized study, they had 15 participants use a removable, denture-like device with real tooth enamel attached, a method developed and extensively tested by Domenick T. Zero of Indiana University, a co-corresponding author on the current paper.

The study participants applied a sugar-containing solution to the appliance four times a day, mimicking high-sugar meals and snacks consumed in the course of daily life. Participants were asked not to brush the enamel specimens but instead to rinse the appliance twice a day. Participants were divided into three groups, with one using the ferumoxytol then the hydrogen peroxide rinse, one with a solution that provides the inactive ingredients in ferumoxytol, and a third with water alone.

After 14 days, the researchers analyzed the biofilms that accumulated on the enamel specimens. They found the experimental treatment potently reduced the growth of biofilms containing S. mutans and could kill this bacteria with high specificity. Other commensal bacteria normally found in the mouth were not affected by the ferumoxytol-hydrogen peroxide therapy.

The results and safety of the approach were supported by previous work, which has shown that the iron oxide nanoparticles do not bind to the mucosal tissue in the mouth and do not cause cytotoxicity or changes in the oral microbiome in an animal model.

?This treatment doesn?t seem to have harmful, off-target effects,? Koo says.

The reason for this precision is three-fold. The reaction to catalyze hydrogen peroxide only happens in a highly acidic environment, such as what arises when caries-causing bacteria are present and active. The treatment also causes the sticky matrix of carbohydrates to break down, degrading the biofilm and exposing the microbes. And ferumoxytol can specifically bind to receptors on the S. mutans cell membrane, facilitating its killing.

In a final stage of the study, the researchers added a marker that turns blue when exposed to reactive oxygen species, like those generated by the catalysis of hydrogen peroxide by ferumoxytol nanoparticles. And indeed, the team found that the intensity of blue labeling corresponded with acidic biofilms containing S. mutans.

Because of this ?detection? capability, Koo notes that the experimental treatment has the potential to become what is known as a ?theranostic,? that is, a drug that can be used to both diagnose a condition and treat it. ?It can also be used at home,? Koo says. ?You could rinse with it, see how much cavity-causing plaque there is, and then treat with the solution or consult a dentist for follow-up treatment.?

Let’s start immediately from the present, your EP Dance Mania Stereo Love is out, Sofia can you present it to our readers of AllMusicItalia?

DANCE MANIA Stereo Love it’s a really special project for me, not only because it’s my first EP, but also because it allowed me to dig deeper into myself. I loved every moment of writing the record, we spent unforgettable moments and I hope it can accompany those who listen to it by letting themselves be overwhelmed by the music.

The new album marks an artistic maturation, a change of sonority that leaves behind more acoustic pieces to a great production of Maestro who is also co-author of the songs. Dance Mania Stereo Love is a pop dance album in which in addition to the sound there is also a particular care for the lyrics, can you deepen this aspect?

Well this album marks a new chapter in my journey, I would just say that I have grown up. I think that music is the mirror of our soul and that it too has changed with me. Maestro played a fundamental role, we were able to immediately get in tune with the lyrics as well as the sound. I don’t like to explain the meaning of my songs, I prefer to leave this margin of freedom to those who listen to them, so that they can give vent to their imagination and let themselves be carried away by emotions.

The song “Solo”, released last year seems to me to be the link between the two styles, the transition from acoustic songs to pop dance. Correct?

Let’s say yes, Solo it was the first song I wrote together with Maestro and that started the realization of the EP.

The first excerpt from the EP is “The Impossible” which has a text focused on an inner path that matures and finds its turning point.
“How beautiful is the world / Things I will never know / You know loneliness / It is what closes in me / even if I never really had you / I see you / I feel you behind everything” until you get there to “How beautiful is the world / No it is not possible to race against time to return to you and tell you how beautiful the world is / Follows the impossible / It is only a word made to bring me to you / And to tell you that I am there still behind everything “
It is a beautiful message that comes from you, what prompted you to write it?

The idea of The impossible came to life during quarantine.
It was a flash inspiration, enclosed in 2 days of study with Maestro ed Eiemgei. I was going through a difficult moment, you know, when everything seemed to collapse on me the song was like a mantra, I hope it can be also for those who listen to it.

The musical part is very present in the songs and at times it seems, at first listening, to dominate the text but it is not so, it is at the service of it. The texts are very accurate and must be listened to well. Are they born first from the textual or musical part? Or do you compose them together?

As far as writing is concerned, it is usually natural for me to start from the musical part, usually it is the melody that comes first. It must be something that stays in my head and already from that sound a word can often be formed and so on. Often they are born together but not necessarily, sometimes even just some keywords are enough to unleash an entire concept even before having started playing.

Personally, the whole EP intrigued me, you catalyzed me in listening. Besides ?The Impossible? I was struck by ?Well so?, can you tell us how these songs are born?

They are both my favorite EP songs. I can’t tell you how or why they are born, often there is no real story behind each song, sometimes it was enough just to make a round of chords or hum a melody to then give life to the songs.

Laotians do not want a giant Chinese Buddha: the Chinese real estate group Wanfeng Real Estate, of Shanghai, which since 2012 has been building a 3.65 square kilometer special economic zone in Vientiane, in the That Luang swamp region, in the east of the Laotian capital, is under fire from critics. In question, the project to erect a giant statue of Buddha, intended to become the central attraction of an aquatic theme park for tourists – mainly Chinese.

The project is not new. But its implementation seems imminent, since the country’s Deputy Prime Minister, Kikeo Khaykhamphithoun, visited the promoter’s premises at the end of August to learn about it. That the communist leader – Laos has been led since 1975 by the Lao People’s Revolutionary Party – welcomed, according to the official Laotian press, that this “Gigantic representation of Buddha can give an impetus to Buddhism in the era of globalization, while encouraging the economic development of the SEZ”, still happening. But that he was introduced a replica of the future statue, in the form of a standing Buddha, in the typical style of the Mahayana school known as the “great vehicle”, the majority in China and North-East Asia, causes a shock: in Laos, where predominates, as in Thailand and in Burma, the Theravada school known as the ?small vehicle?, the Awakened one – the meaning of ?Buddha? in Sanskrit – is most often represented in a sitting position of meditation.

Critics immediately rocketed on social networks. “They should build a Laotian Buddha, or if they want to build it as a symbol of cooperation between the two countries, then they build two Buddha statues – one Chinese and one Laotian side by side., a Vientiane resident told Radio Free Asia’s Laotian service. If they are content to build only that of China, Laos will lose face ».

“Symbol of sovereignty”

For Adisorn Semyaem, director of the Mekong Studies Center at Chulalongkorn University in Bangkok, quoted by Nikkei Asia magazine, “It would be unacceptable for Laotians for a large statue to be built in the That Luang Swamp Special Economic Zone to attract tourism, not far from the Pha That Luang stupa, the symbol of Laotian sovereignty.”

In reality, the distinction between the two schools of Buddhism is far from being so clear: there are many statues of seated Buddhas in China. And at least one dispatch from the Laotian press agency quoted, in August 2020, that is to say a year earlier, a Chinese official evoking a “Statue in a seated position” of a hundred meters for the Chinese economic zone. The question, according to connoisseurs, has more to do with Laotian nationalism, faced with a China which is multiplying infrastructure projects in this small country of 7.5 million inhabitants with which it shares a border.
