Having at least some omega-3s in red blood cells was associated with better brain structure and cognitive function among healthy study volunteers in their 40s and 50s, according to research published online Oct. 5 in Neurology®, the medical journal of the American Academy of Neurology. Faculty of The University of Texas Health Science Center at San Antonio (UT Health San Antonio) and other investigators of the Framingham Heart Study conducted the analysis.

?Studies have looked at this association in older populations. The new contribution here is that, even at younger ages, if you have a diet that includes some omega-3 fatty acids, you are already protecting your brain for most of the indicators of brain aging that we see at middle age,? said Claudia Satizabal, PhD, assistant professor of population health sciences with the Glenn Biggs Institute for Alzheimer?s and Neurodegenerative Diseases at UT Health San Antonio. Satizabal is the lead author of the study.

Volunteers? average age was 46. The team looked at the relation of red blood cell omega-3 fatty acid concentrations with MRI and cognitive markers of brain aging. Researchers also studied the effect of omega-3 red blood cell concentrations in volunteers who carried APOE4, a genetic variation linked to higher risk of Alzheimer?s disease.

The study of 2,183 dementia- and stroke-free participants found that:

Researchers used a technique called gas chromatography to measure docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) concentrations from red blood cells. The omega-3 index was calculated as DHA plus EPA.

?Omega-3 fatty acids such as EPA and DHA are key micronutrients that enhance and protect the brain,? said study coauthor Debora Melo van Lent, PhD, postdoctoral research fellow at the Biggs Institute. ?Our study is one of the first to observe this effect in a younger population. More studies in this age group are needed.?

The team divided participants into those who had very little omega-3 red blood cell concentration and those who had at least a little and more. ?We saw the worst outcomes in the people who had the lowest consumption of omega-3s,? Satizabal said. ?So, that is something interesting. Although the more omega-3 the more benefits for the brain, you just need to eat some to see benefits.?

Researchers don?t know how DHA and EPA protect the brain. One theory is that, because those fatty acids are needed in the membrane of neurons, when they are replaced with other types of fatty acids, that?s when neurons (nerve cells) become unstable. Another explanation may have to deal with the anti-inflammatory properties of DHA and EPA. ?It?s complex. This new study addressed these important research questions, finding strong and linear associations of depression across all personality traits.?

Among the study?s findings was that people with high agreeableness were 49 percent less likely to become depressed than people with low agreeableness. Additionally, those with high neuroticism were twice as likely to develop depression than those with low neuroticism when using more than 300 minutes of social media per day. More importantly, for each personality trait, social media use was strongly associated with the development of depression.

The sample of more than 1,000 U.S. adults between the ages of 18 to 30 was from 2018 data collected by Primack and his colleagues at the University of Pittsburgh.

Depression was measured using the Patient Health Questionnaire. Social media was measured by asking participants how much daily time was spent using popular social media platforms, and personality was measured using the Big Five Inventory, which assessed openness, conscientiousness, extraversion, agreeableness and neuroticism.

The authors suggest that problematic social comparison can enhance negative feelings of oneself and others, which could explain how risk of depression increases with increased social media use. Engaging primarily in negative content can also enhance these feelings. And lastly, engaging in more social media reduces opportunities for in-person interactions and activities outside of the home.

Depression has been noted as the leading cause of disability and mortality worldwide. This makes these findings even more pronounced for creating health interventions and prevention efforts.

?Findings from this study are important during a time of technology expansion and integration,? Merrill said. ?Connecting to people virtually may increase the risk of miscommunication or misperception that leads to relationship difficulties and potential risk for developing mental health problems.?

?People have innate emotional needs for social connection and understanding,? Merrill added. ?For example, social media experiences can be improved by becoming more aware of our emotions and our connection with others in various life circumstances. This awareness helps improve relationship quality by simply reaching shared meaning and understanding through more effective communication and concern for others and ourselves. ?Cystic fibrosis is a genetic disease that is associated with increased inflammation, and like many inflammatory diseases, it comes with a large amount of oxidative stress,? said Maret Traber of OSU?s Linus Pauling Institute, who led the study.

Oxidative stress happens because of an imbalance in the body between free radicals, which can cause harmful chemical reactions, and antioxidants. Antioxidants are molecules able to give an electron to a free radical, causing the free radical to become less reactive without becoming unstable themselves.

?In addition to oxidative stress, cystic fibrosis is also characterized by problems with fat absorption, which limits uptake of vitamin E, a fat-soluble antioxidant,? said Traber, a professor in the Oregon State College of Public Health and Human Sciences. ?Low vitamin E levels plus high oxidative stress is a recipe for more inflammation, which can contribute to a range of negative health outcomes.?

Patients? difficulties with fat absorption mean that they need to consume larger than usual amounts of fat-soluble vitamins like vitamin E, Traber said. Patients need at least 400 milligrams of vitamin E daily to achieve normal blood concentrations, she said.

In this study, the researchers looked at whether vitamin C supplements could help patients better use their absorbed vitamin E. Vitamin C can recycle oxidized forms of vitamin E, and it is also helpful at tamping down the oxidative stress aspects of inflammation, Traber said.

After 3½ weeks of daily 1,000-milligram doses of vitamin C, the patients in the study trended toward lower blood concentrations of a key oxidative stress biomarker, malondialdehyde or MDA, and also toward a slowdown in vitamin E elimination from the bloodstream.

?Since vitamin E is hanging around longer, it might be able to get into tissues better, and better protect cell membranes from oxidative stress,? Traber said.

The benefits of the findings, published in the journal Nutrients, are not unique to cystic fibrosis patients, she noted. Smokers, for example, typically have problems associated with oxidative stress and can benefit from extra vitamin C and possibly extra vitamin E. Metabolic syndrome patients have issues with vitamin C and E as well.
